The man, aged in his late teens, was winched to safety shortly before 10am after suffering suspected leg and pelvis injuries.
He is now being taken by road ambulance to The Alfred Hospital in a stable condition and with serious injuries to his legs and pelvis.
The Metropolitan Fire Brigade said the rescue operation was delicate and involved the use of ariel appliances, with crews expected to be at the scene for some time.
A MFB spokesman said more than 20 firefighters were on the scene.
“Rescuing the man is a delicate operation and crews are expected to be on scene for some time,” he said.
It’s understood the man fell down into the drain at a construction site.
A caller told 3AW the property had a ‘demolition site’ sign on the front and that there was a heavy police presence.
“There were at least six fire engines, one paramedic van parked around the corner,” caller Bernadette said.
“Police cars and police absolutely everywhere and a chopper hovering over head.”
Worksafe has confirmed they will investigate.
