The blue Mitsubishi ASX, with a West Australian registration plate 1HET006, was found crashed on the Eyre Highway at Mudamuckla, about 35km east of Ceduna, at 4.20am Wednesday.
Patrols had spoken to the occupants of the car about 3.30pm the day prior at a car wash on Kuhlmann St in Ceduna.
One of the missing occupants is described as a woman aged about 20, with black hair and pale skin who was last seen wearing black shorts and an Adidas crop top.
The man she was travelling with is described as being aged 39, with a stocky build, facial hair and a gold chain.

It comes after the body of Dylan Stuart was found in the state’s Far North earlier this month.
Mr Stuart, 29, and his partner Danika Thomson, 24, became bogged in their 4WD near Junction Dam on the Stuart Creek Station, about 90km north of Roxby Downs on February 26.
Emergency services crews were alerted to the missing pair the following day after family could not contact them.
A large scale search was launched and Ms Thomson, 24, was located on Monday, February 28, along the creek line about 100km north of Roxby Downs.
She had left their vehicle on foot, after Mr Stuart had not returned to the 4WD for several hours the day they became bogged despite leaving on a motorbike to seek help.
Ms Thomson was treated for dehydration, and Mr Stuart’s body was sadly located on March 1 near Gregory Creek, about 100km north of Roxby Downs.
