Segeyaro will need surgery after he suffered an ankle syndesmosis injury in the Cowboys' 28-12 loss to the Gold Coast Titans on Friday night at Skilled Park.
The injury occurred during the first half when he twisted his ankle awkwardly.
The diagnosis is a 14 week lay-off at best.
Segeyaro has been one of the finds of the NRL this season, with his excellent form earning him a rare run-on start against the Titans.
The injury will also cloud his attempts to negotiate a new contract with the Cowboys, with several clubs also interested.
