Merri Parade has been closed inbound between St. Georges Rd and South Park St in Northcote after the truck smashed into the railway bridge about 8.10 am on Friday.
Police are directing traffic onto Charles St.
Paramedics treated two people at the scene but were not taken to the hospital.
Authorities are warning motorists to allow extra time on local roads around the area and consider using High St instead.
It comes one day after the Burnley Tunnel was brought to a standstill after a Coles-branded truck triggered height sensors but continued to drive through.
