Matt Doric claimed the key scalps of Ramneet Dhindsa (4) and Kingston Hawthorn captain Ryan Ninan (45) on his way to a haul of 4-66 from 21.2 overs.
Doric also had Justin Kramersh (35) and Sacheen Paramesh (0) caught behind, his 10th and 11th wickets of the campaign.
The Hawks had navigated their way to 2-81, but when opener Jake Marnie (33) was run out by Aaron Ayre, Essendon sensed an opening.
James Seymour (2-25) trapped Akshay Kodoth lbw for 43 and when Shorye Chopra fell to Nishal Perera (2-34) Kingston Hawthorn had lost 3-18 to fall to 5-99.
Fighting lower-order hands from Jack Fowler (22) and Kramersh enabled the visitors to pass 200 at Windy Hill.
While Seymour perished for 48 with stumps in sight, he put Essendon in the box seat alongside Ayre (33 not out).
The pair took the attack to the Hawks bowlers in 19 overs before stumps, with Ayre and Isaac Conway (4 not out) to resume on Saturday.
Dishan Malalasekera (1-19) was Kingston Hawthorn’s wicket-taker, trapping Seymour lbw.
Essendon is striving to cement itself in the top eight, while the Hawks will be chasing their first victory of 2018-19 on day two.
