Police said the male driver of the Mitsubishi — and sole occupant — died at the scene. He is yet to be formally identified.
The male driver of the Range Rover, and his two passengers, a man and woman, were released after being trapped for a short time.
The woman was flown to Westmead Hospital in a critical condition while the men were taken to John Hunter Hospital with serious injuries.
Officers from Tuggerah Lakes Police District have established a crime scene and will investigate the circumstances surrounding the crash.
The road has been closed in both directions with Red Bus cancelling all number 29 bus services along Wilfred Barrett Drive until further notice.
A report will be prepared for the Coroner.
