Bolton replaces co-captain Matthew Scott who has a broken finger.
The Cowboys are chasing a sixth straight victory and need to win to qualify for the top eight.
Bolton last started at prop in round 12 against the Gold Coast Titans at Skilled Park when Scott and James Tamou were on Origin duties.
With Bolton promoted to start, Ashton Sims returns to the interchange bench for his first game since round 23 when he copped a one-match suspension against the Titans.
Matt Bowen will run out in front of his home crowd for the last time.

The Titans have named an unchanged 18-man squad for Saturday night's clash with Storm in Melbourne.
Coach John Cartwright has named co-captain Nate Myles in the side in the hope he will overcome an ankle injury suffered during last Sunday's upset win over the Sydney Roosters.
Myles has been cleared of serious ankle damage and will attempt to prove his fitness later on in the week when the team travels to Melbourne.
Prop Mark Ioane has again been named as 18th man on an extended five-man interchange bench.
